Preparation of culture medium
1. Preparing the media:
Generally prepared culture medium one week in advance, to check that containers have not been contaminated during preparation. The preparation is the same, regardless of the chosen medium.
It's like a recipe:
a) In a large pot, pour 1 liter of distilled water, deionized water or osmosis water. Heat until the water boils. Then turn down the heat, to stop the boil but keep good hot water (simmering)
b) Caution: If the medium already contains agar, skip to step c.
Agar pour into the pan and stir with a whisk periodically until the mixture again become clear (approximately 10 minutes). It takes a dose of 7 grams of agar for 1L culture medium.
c) Pour carefully the media bag in the pan, because the powder is very volatile and could spread easily in the event of inadequate gesture.
d) Allow to heat without boiling, whisking regularly for 10 to 15 minutes at least.
e) Remove from heat and pour the medium into your tuperware, about 1cm thick per container (with a syringe, it's easier)
f) Drill the covers of a 1mm hole diameter and place a micropore tape piece. Put the lid when completely cooled.
2. Sterilization of tuperwares:
a) tuperwares are placed in the basket steam pressure cooker in which one has previously paid a water depth of approximately 1 cm.
b) Close the pressure cooker and put it there on high heat
c) When the pressure cooker starts to whistle, lower the heat so that the valve whispers gently and count then 24 minutes 'cooking'
d) The 24 minutes are up, turn off the heat (or the casserole out of the plate if your stove is electric) fall completely and let the pressure cooker and cool completely before opening
e) Take them out of the pressure cooker. On cooling, the agar solidifies. Shoot them with cellophane so as not to allow air to enter. They can be twisted out of the pressure cooker but will resume their original shape after cooling.
After two hours, store your containers at room temperature, protected from shocks (buffet, shelves, etc.) can also be stored in the refrigerator if you do not wish to use them for several weeks.
f) After three or four days, check them for possible contamination. Discard those that you consider questionable (whitish deposit, moss, etc. ...)

Sterilization of Microbox
Sterilization of these boxes will be with a wick placed in a corner and closed lid the other 3 corners. Place the boxes and start the fire so that the pressure rises slowly.
After 25 minutes of cooking, let the pressure down and Ouveze your casserole.
Remove the strip and close the last corner.
